KinomeScout™

Target profiling for kinases and other ATP-binding proteins

Submit a project inquiry

KinomeScout™ is a versatile and well-established tool for kinome profiling that works with native proteins under physiological conditions to direct the design of potent and selective small-molecule drug candidates, while circumventing typical downsides of protein kinase panel assays. The technology is based on proprietary kinase-binding matrices that can be used to profile over 300 different kinases and other ATP binding proteins (such as helicases or phospho-diesterases). Thanks to our Kinobeads-like chemical proteomics approach, kinase inhibitors can be profiled without the need for labels on compounds or proteins. The KinomeScout™ technology enables selectivity profiling by testing hits, leads and drug candidates against all kinases in a tissue or cell type in a single experiment, facilitating identification of off-targets and providing physiologically relevant selectivity and affinity information (such as EC50 and estimated Kd values).

We offer multiple workflow options to match your experimental needs. The assay is also readily available as KinomeScout™ HT format for profiling 1,000 to 10,000s of protein kinase inhibitors in a reasonable timeframe.

We can also profile kinase inhibitors and other ATP-binding proteins using activity-based profiling approaches, such as ActivX (KiNative™).

Frequently Asked Questions

Is a compound label required for KinomeScout™?

No – KinomeScout™ leverages a Kinobeads-like chemical proteomics approach for kinase inhibitor profiling, avoiding the need for compound labeling.

How many kinases can be profiled by KinomeScout™?

Momentum has proprietary knowledge of various affinity matrices to profile >300 kinases, including 42 understudied protein kinases (‘dark kinases’).

What is the throughput of KinomeScout™?

Our traditional KinomeScout™ workflow is a low-throughput approach for the profiling of one to several compounds of interest. Our high-throughput KinomeScout™ offering – KinomeScout™-HT – enables 1,000 to 10,000s of protein kinase inhibitors to be profiled in a reasonable time frame. In one previous client project, KinomeScout™-HT was used to profile 1000 compounds at four concentrations within three months.

Can KinomeScout™ only be used to study kinases?

No – the KinomeScout™ workflow is compatible with other non-kinase ATP-binding proteins. If you have questions about your target's compatibility with KinomeScout™, contact our scientific team.

Does Momentum offer other target profiling approaches?

Yes – Momentum offers a range of target profiling technologies to best serve your specific experimental needs. Alternative approaches include TargetScout™, PhotoTargetScout™, CysScout™, SPICE, and CHIPP.
