Radiotherapeutics represent a powerful clinical modality, particularly for the diagnosis and treatment of cancer. However, limitations and restrictions associated with the laboratory use of radioactivity can make radiotherapeutics drug discovery and development uniquely challenging. At Momentum, we upgrade the traditional approach to radiotherapeutic development by coupling the use of a surrogate payload with the ultra-sensitive quantitative detection capabilities of ICP-MS. As a result, radioligand candidates can be identified and characterized safely, simply, and rapidly to support the discovery and development of effective radiotherapeutics.

Our radiotherapeutics discovery and development strategy avoids the hazards and restrictions associated with radioactivity by using a lanthanide or other surrogate payload during compound validation, optimization, and characterization. The behavior of these surrogates in laboratory testing enables radiotherapeutic activity to be extrapolated prior to integration of the final radioactive payload, accelerating discovery efforts and significantly reducing the barriers to radiotherapeutic development.

At Momentum, we leverage Affinity Selection Mass Spectrometry (ASMS) for the high-throughput screening of ligand candidates. Once a promising lead has been identified and validated, it can be paired with a surrogate payload of choice and further characterized using ICP-MS and other methods.

Affinity Selection Mass Spectrometry
Affinity selection mass spectrometry (ASMS) is used to identify small molecule ligands against targets (including membrane proteins, protein complexes, and DNA/RNA). After validation and characterization, ligands identified by ASMS can be further developed into powerful therapeutics.
ICP-MS
Inductively Coupled Plasma Mass Spectrometry (ICP-MS) can be used to quantify surrogate lanthanides with parts-per-trillion sensitivity from complex matrices (cell pellets, tissues, biofluids). No chromatography is required, and throughput can be further increased by multiplexing.
