PhotoTargetScout™ is a chemoproteomic technology for target deconvolution, selectivity profiling, and binding site identification. This workflow uses photoaffinity labeling (PAL) to directly map interactions between small molecules and proteins in intact cells or cell lysates. A trifunctional PAL probe combines the small molecule of interest with a photoreactive group and an alkyne group. The photoreactive group forms a covalent bond with neighboring amino acid side chains upon light exposure, while the alkyne group facilitates fluorescent labeling or click chemistry-based enrichment of the target. As a result, PhotoTargetScout™ is particularly effective at identifying drug:target interactions in cells, including interactions that are difficult to detect via other approaches. This powerful photoaffinity labeling assay provides site-specific spatial information to enable mapping of the binding interface and is an invaluable tool for the comprehensive identification of on- and off-targets while preserving biological context.
